Oxaliplatin

Oxaliplatin is a platinum-based chemotherapy agent primarily used in combination with fluorouracil and leucovorin (FOLFOX regimen) for the treatment of colorectal cancer. Oxaliplatin exerts its cytotoxic effects by forming inter- and intra-strand crosslinks with DNA, primarily at guanine residues. Oxaliplatin undergoes rapid and extensive nonenzymatic biotransformation in the bloodstream, producing multiple platinum-containing derivatives including cytotoxic species. Oxaliplatin is supplied as a crystalline solid and is available as both an API for pharmaceutical manufacturing and as an analytical reference standard. The compound is freely soluble in water and is typically formulated as an aqueous solution for intravenous administration.

Synthetic Route

An aqueous solution of 5 g of trans-1,2-diaminocyclohexane and 18 g of K₂(PtCl₄) is reacted at room temperature for 12 h to obtain 12 g of intermediate (I). 6.8 g of silver nitrate is added to an aqueous solution of 3 g of compound (I), stirred in the dark for 2-3 h, then 4.8 g of dipotassium oxalate is added, and the mixture is reacted at room temperature for 8 h to obtain oxaliplatin.

Storage Conditions

Store Oxaliplatin at 2–8 °C in a tightly sealed container protected from light. Keep in a dry, well-ventilated area. The compound should be protected from moisture and strong oxidizing agents.
