Daraxonrasib (RMC-6236) is an oral, RAS(ON) multiselective, tri-complex inhibitor of guanosine triphosphate–bound mutant and wild-type RAS protein isoforms developed by Revolution Medicines. It acts as a molecular glue, using cyclophilin A (CypA) as a chaperone to form a tri-complex with RAS-GTP, thereby blocking RAS from activating RAF and inhibiting downstream MAPK signaling .
The compound is a macrocyclic molecule (bRo5) with the molecular formula C₄₄H₅₈N₈O₅S and a molecular weight of 811.05, containing five defined chiral centers . In the Phase 1/2 RMC-6236-001 trial (NCT05379985), daraxonrasib demonstrated objective responses in more than 30% of patients with previously treated RAS-mutant non-small cell lung cancer (NSCLC), with durable responses across dose levels .
