Synthetic Route

A mixture of acetamidophenylsulfonyl chloride (1.0 mmol) and cycloamino (1.5 mmol) in water (1 mL) was stirred for 5 minutes, and the resulting precipitate was collected by filtration. The precipitate was transferred to a round-bottom flask and treated with 2 mL of a concentrated solution of hydrochloric acid and 4 mL of water. The reaction mixture was gently boiled at 100 °C for one hour. The solution obtained from the ice bath was cooled and then alkalized using a saturated aqueous solution of NaHCO₃. The granular precipitate was filtered, washed with water, and dried in air to yield the product, sulfadimidine.

 


Application

Sulfamethoxazole is a broad-spectrum bacteriostatic agent; its active component, para-aminobenzoic acid (PABA), competes with PABA for binding to bacterial folate synthase, thereby inhibiting the synthesis of bacterial folate from PABA as a raw material. This reduces the amount of metabolically active tetrahydrofolate, which is an essential compound for bacterial synthesis of purines, thymidine monophosphate, and deoxyribonucleic acid (DNA). Consequently, sulfamethoxazole inhibits bacterial growth and reproduction. The effect of sulfamethoxazole can be antagonized by PABA and its derivatives (e.g., procaine, tetracaine); additionally, the presence of pus or tissue decomposition products also exerts an antagonistic effect, as these substances can supply the essential nutrients required for bacterial growth.
