Brivaracetam

Model:357336-20-0
Brivaracetam (CAS 357336-20-0) is a potent, orally active anticonvulsant agent belonging to the racetam class of medications, developed as a chemical analog of levetiracetam with significantly enhanced pharmacological properties. Structurally, Brivaracetam is a gamma-lactam and a non-proteinogenic amino acid derivative, specifically (2S)-2-[(4R)-2-oxo-4-propylpyrrolidin-1-yl]butanamide.

Brivaracetam is a pivotal antiepileptic API and reference standard in the pharmaceutical industry, distinguished by its remarkably high affinity for the SV2A target. The superior binding affinity of Brivaracetam 15 to 30 times that of levetiracetam translates into enhanced clinical efficacy at lower doses, making it a valuable therapeutic option for patients with partial-onset seizures. In quality control and regulatory contexts, Brivaracetam is supplied with comprehensive characterization data for analytical method development, method validation (AMV), and quality control applications. The unique gamma-lactam scaffold of Brivaracetam continues to serve as a template for medicinal chemistry research exploring novel anticonvulsant agents and SV2A-targeting therapeutics.

Indication

Brivaracetam is a highly selective and affinity-rich ligand for synaptic vesicle protein 2A, approved for the adjunctive treatment of partial seizures in adult and adolescent patients aged 16 years or older with epilepsy, with or without secondary generalized seizures.

Main Function

Brivaracetam exhibits high affinity for selective binding to synaptic vesicle protein 2A (SV2A), the action site of the antiepileptic drug levetiracetam. SV2A is located on the presynaptic membrane and participates in regulating neurotransmitter release and vesicle recycling, thereby maintaining normal synaptic vesicle function. Binding of brivaracetam to SV2A reduces the release of excitatory neurotransmitters and controls epileptic seizures by modulating the balance between excitatory and inhibitory neurotransmitters in the brain. The affinity of brivaracetam is 1530 times that of levetiracetam, resulting in a approximately 10-fold reduction in required dosage.

Application

Brivaracetam is a next-generation antiepileptic drug approved by the FDA for the treatment of focal seizures in patients aged 16 years and older, as well as for use in laboratory research and pharmaceutical development processes.
