Lifitegrast is a leukocyte function antigen 1 (LFA-1) antagonist used for the treatment of dry eye syndrome. Lifitegras binds to LFA-1, preventing its interaction with its homologous ligand intercellular adhesion molecule-1 (ICAM-1). ICAM-1 is overexpressed in the corneal and conjunctival tissues of patients with dry eye disease; the binding of LFA-1 to ICAM-1 promotes the formation of immune synapses, thereby facilitating T-cell activation and migration to target tissues. In vitro studies have demonstrated that Lifitegrast inhibits T-cell adhesion to ICAM-1 and suppresses the secretion of inflammatory cytokines in human peripheral blood monocytes.
